These elements help the most with this bit- the progression of the people in the diner realizing what's happening and being powerless to stop it. Mulaney has a very good way of emphasizing words and ideas that make his story telling punchier and feed into the central joke of the bit. The situation itself is hilarious too- no one expects to play "What's New Pussycat" 20 times with a "It's Not Unusual" thrown in for good measure.

Situation and expectation come in to play as to why this scene is so funny. The thick lisp creating a communications barrier, and the expectation (and anticipation) of the centurions finally breaking into laughter aids the second leg of the bit.
What really makes this for me is the fact that the centurions came from a actor's guild not affiliated with the Pythons, and they were told the absolutely could not laugh, for time's sake. Their reactions are genuinely of people trying desperately to keep their job, since they believed they'd be fired for laughing.
